Billy and Jimmy Lee (Double Dragon)
When it comes to beat-em-ups, for many people, Double Dragon is the signature series. Of series in that genre, it certainly had the most outside media presence, with a movie, a cartoon, and other endeavors, as well as a fighting game spin-off, a crossover with the Battletoads, and several rereleases and revival games in somewhat recent years, such as Double Dragon Neon and Double Dragon IV, as well as cameos and crossovers with the River City series' own revivals. I pick the Lee Brothers over Kunio-Kun just about because they are likely more known in the US and Europe, as big as Kunio-Kun is/was in Japan.
The two brothers have no problem fighting, given that they fight a big street gang (and sometimes each other) all the time. They can do a Whirlwind Kick that propels them in the air, shoot out a spiraling energy beam of two dragon heads (Double Dragon, go figure), and even shoot forward with dragon energy that engulfs their whole body. But beyond energy attacks and various brawling moves, you can also do unique things with them, mechanically. You could either have a duo or Jimmy as an alt for Billy. For a duo attack, you can have them High-Five to buff each other in various ways. You could have them find and pick up various weapons, or have it as a move that has them switch weapons [hand to hand -> knife -> chain -> baseball bat -> trash can, etc.]. You can have them also use the tapes from Double Dragon Neon for even more buffs, as well as specialized moves like fireballs, bombs, lightning bolts, and dragon spirits. Basically, you can figure out many ways to make them more than "just" brawlers.
