For appearances in Mario games...?I you haven't noticed, Funky appeared in MK Wii and Super Mario Sluggers, while K. Rool is 1 for two. Funky could also be an easy last-minute semi-clone and has such an awesome attitude. Prepare your anus, Diddy, the Funky Train is here!

I am going Left to Right while listing the newcomers.
Please keep in mind my former statements regarding Sakurai's outlook in adding newcomers, and how his main goal is unique characters with moves no one has before.
Diskun - He kind of functions as a placeholder for a "WTF Retro" that we have seen in Melee in the form of Mr. Game & Watch, and in Brawl in the form of R.O.B. I would like is Diskun fit this role, which is why I put him down as the placeholder for this.
Rosalina & Luma - Confirmed playable in the Nintendo Direct on 12/18/13.
Dixie (in a Diddy tag-team) - I feel that Dixie deserves a spot on the roster, but she is not that different from Diddy Kong moveset wise. It would cool if Sakurai put here on an Ice Climbers-like team with Diddy, were they would swap places as leader and follower by using a move (probably Down B, and then some of their moves would change.
King K. Rool - Even though he is not recent, he would make a very cool addition. He is a crocodile (Kremlin to be correct) and can also be a pirate, scientist, or even a boxer! This is definitely a unique newcomer for Smash, and none of these titles can fit into another character (except for Little Mac as a boxer, and maybe Tetra as a Pirate).
Tingle - This one is a little tricky, but he is definitely a uniqe character. He is hated over here in the West, which may decrease his chances, but he has his own spin-off series and is popular in the East in Japan, which boost his chances. He could use Balloons as Villager seems to do and Balloon Fighter would have. He could use Radom items from the Zelda series, including rupees, bottles, or even maps somehow!
Ridley - I'm starting to get a little but worried about Ridley here. I am not worried about the size issue at all, it's Sakurai's hints towards him being a stage hazard that worry me. Though if he was in, he could have a lot of potential and new moves to offer, given that he is an alien dragon thing.
Takamaru - I am not so sure about this one uniqueness wise. We don't have any sword samurai/ninjas in smash yet, so he could be unique, but we do have two Blue haired swordsman.
But Takamaru could be unique in his own way.
Vince - As Rosalina & Luma are the new Super Mario chracters, it seems that we won't be getting Bowser Jr. this time around in smash. This is a bit weird as Sakurai seems to e looking for unique characters, and Bowser Jr. using his paintbrush seems to be VERY unique! I have a feeling Sakurai acknowledged Jr.'s paintbrush abilities, and wanted to assign it to someone else so Rosalina could take the 5th Mario character slot. The most likely choice for paintbrush moves would be Ince from Art Academy, as it is a new and loved series. Other art characters could be Adeline or Smeargle, but Adeline only appeared in ONE game (possibly 2) in the Kirby franchise as awesome as she may be, and Smeargle is just a random Pokémon as awesome as he may be.
Mewtwo (returning from Melee) - Pretty obvious choice as he has his new X and Y forms, which he could implement in a fun play style somehow. My only concern is that he would need new special moves just to differentiate him from Lucario more, even though they are not completely the same.
Roy (returning from Melee) - Though he is not very likely, it's the fan demand that would bring him in. He could be more unique if he moveset is changed a little so his fire sword attacks are used in a new and unique way that differentiates him from Marth and Ike.
Villager - Confirmed in the E3 Nintendo Direct on 6/11/13
Mii - This all depends on if customizability plays a role in him/her, and if so how will it be used. If he/she is just a Mii you choose from the console to play as, and has an already set moveset, this can still be a cool character. He/she can use sports attacks (the only other one who can do this is Waluigi, who is very unlikely do to Rosalina & Luma, though he has a very small chance of appearing as a Wario newcomer), vehicles from Pilotwings Resort, or other things from other "Wii _____" games.
Wii Fit Trainer - Confirmed by Mashahiro Sakurai himself at E3, a little while after the E3 Direct on 6/11/13.
Little Mac - A very likely character, but his uniqueness depends on how he uses his boxer punches. I'd he has a star punch gimmick for countered attacks, that would be very unique! It all depends how Sakurai does him. Can be good or bad.
Reggie - Nintendo seems to be advertising Reggie a lot these days, with making a "My Body is Ready," joke whenever they have that chance. Just watch that last direct. There was a freaking Reggie Mii walking around! And he did an arm wrestle with real Reggie at the end, to which Reggie won, which was probably a joke to how his body was ready or soemthing. Even Reggie said he wnats himself in Smash Bros! He even tweeted, "My #finalsmash would be the "MY BODY IS READY SLAM!"
I mean come on Reggie! COME ON!
Mega Man - Confirmed in E3 Nintendo Direct on 6/11/13
Professor Layton - He functions as a "half placeholder" on my roster. I think Smash 4 will have a 4th 3rd party charcter on top of Sonic, Snake, and Mega Man. This will probably be either Pac-Man, another Namco-Bandai character like Lloyd Irving, a Square-Enix character (probably Slime or DQ hero), or Profesor Layton. Out of all of these, I would prefer Layton, with Slime in a close second. And I think Layton does have a solid, standing chance.
Chibi - Robo - Though I never actually played the original GameCube Game, I have seen gameplay, and the upcoming 3DS game looks pretty good! He could become the smallest character on the whole roster, being an inch shorter than Squirtle. I am not sure about his moveset, as I said I never aged the original game, but I know for a fact he can be VERY unique!

You know Diddy clone, he's as serious as a heart attack.I want Waddle Dee in a lot! It's just Sakurai's modesty that concerns me.
It confuses me how he is modest and doesn't throw in his own characters, but he goes if he along and trolls the Kirby Air Ride fan base, the MarioKart 8 fan base, and people who don't want Rosalina aim smash bros all in one 2 and a half minute video.
And I like how everyone likes the idea of 50 charcters and says it is the perfect roster number, but if I put in 2 more characters everyone says it is comeptlely out of the question.
But seriously.....
I would replace Isaac or a third party with Daitouryou. That is what I recommend.

Regardless of how I feel about Dr. Luigi, I can't even begin to deny Mr. L.I refuse to take Dr. Luigi seriously. He doesn't take off his hat, nor does the man have a catchy theme song. How else will he throw brightly colored pills? Besides, Mr. L is the best Luigi alt.
