It's confirmed not to be a fake account, it could be a joke since layton says: "I was told to fight! ................................................ im leaving!!

But it is nintendo's official layton account "tophatprofessor"

Like I said, it was written by an employee of Level-5 as a joke, it was a suggestion, though if Level-5 does indeed get in contact with Mr. Sakurai we could very much see Layton in the game but that's if it were to happen, otherwise he seems pretty unlikely but not impossible.
