I would probably say at least some of the stages will be playable.
Really? I was sure we were gonna fight on a platformless backdrop, and everyone gets KO'd the second the match starts.

Anyway, I would say Battlefield, Wily's Castle, Boxing Ring, and Palutena's Temple.

Boxing Ring maybe (that's probably why the secret light platform was revealed recently, so you know about when you go to best buy).
Battlefield and Final Destination most likely.
Maybe Wiley's Castle.

Hmmm.... Final Destination and Battlefield seem like obvious choices. Wily's Castle perhaps? Past stages aren't likely. I expect most announced stages minus Pyrosphere.

Well, if they do the Final Destination versions of the stages I could see them all being playable. Though, I doubt Palutena's temple has a an FD version.

They're probably going to include a good amount of the gimmicky levels too. Those seem to be the ones that attract kids more.

Stages? I'm sure there wouldn't add Pyrosphere unless his fate is decided at E3.
FD is a given, probably going to be the final stage they are going to fight in.
And it seems like a bet that there would be home stages to newcomers, i.e Town and City, Dr Wily's Castle, Wii Fit Gym etcetera.

Reggie's face must be playable in the demo! If not, I'll be disappointed. If it's not in the game at all, I'll be ultra disappointed, but I'll still get the game either way.

I really hope they include the Pilotwings stage. If not, at least have Skyloft.

They'll probably include the stages that move so that the WiiU's power/graphics are showcased better. So probably Skyloft, Pilotwings, and maybe the boxing ring as well.

Why not Pyrosphere?
In my case, it's a little bit of a joke (that's intended a small, not really funny jest) about how Sakurai has essentially, at least in some people's eyes, been dancing around confirming/deconfirming Ridley as a playable character. The reveal during the direct that Ridley would make some sort of appearance on the Pyrosphere. Essentially, the joke is that Sakurai being Sakurai, will not allow the demo to have the Pyrosphere, thus preventing us from knowing if Ridley is indeed a boss like was implied or if Sakurai was just teasing/trolling.

What this guy said, except it's not a joke. I could legitimately see there being no Pyrosphere, just for the sake of prolonging this Ridley s*** even longer. And it'll change nothing, because the arguments would just become either "What are they hiding by not showing the Pyrosphere?" and "Oh, they're still, still, still working on Boss Ridley or hiding him as a fun surprise!"
