Most likely, the wouldn't spoil the characters if they weren't.
Sonic and Snake are known to be unlockables. Sakurai noted in his journal that all third-party characters would be such, due to being "guest" characters in the game.
Beyond them, its anybody's guess at this point. Personally, I suspect that the newcomers from Metaknight on are unlockables, and that Sakurai revealing starters before unlockables was what caused Metaknight's delay. But we won't know until late January.
