Dr. Mario is NOT going to be in Brawl. He was the first to go. He was a clone and it was pointless to include him to begin with. They could have given Melee another character and dropped Doc. Like... Dedede.

And yeah, Doc is probably gone. He has no reason to be in any longer - he was only in Melee because Sakurai liked his music, and the new music mode makes him no longer required for that - and is terribly redundant with Mario already in. The best you can hope for is for him to be an alternate costume of Mario.

But we know for a fact some are leaving. Read [thread=72642]the translations of Sakurai's journal[/thread], where we get this piece of info (specifically, entry #81-90).

General opinion among the smash community is that Doc, Pichu, and Roy are definitely out (Doc and Pichu due to redundancy, Roy due to being an add in Melee and being fairly poor as an overall FE rep). There are arguments over the return of Young Link, Sheik, and Falco; and to a lesser degree the others with the exceptions of Luigi, Captain Falcon, and Ganondorf, who seem to be the only ones almost universally agreed to be returning.

Those are the ones that I'm highly interested it although narrowing them down is tough because whenever I play a new smash I just go into vs mode then play spasticly with one character for 2 minitues until I have played all the characters... then I usually just pick one the go on a unlocking rampage.
