I would love to see Serynder or Master Puffy have a go at the new Puff. I don't think there really is a prominent puff main in japan.

She retains the same costumes from Smash 4. Which is weird because Pikachu got new female costumes (as opposed to the all male ones it had before) but Jiggs still has all the old feminine costumes (aside from the gender neutral Green Sleep Cap)

Puff getting ignored by the development team for changes is on par for the course at this point. They think her being in the game is enough to please fans. She's easily the most neglected out of the original 12. you always see the other original 12 in some kind of promo for the new game but...

King K Rool got a mii costume and made it in. I'm sure Sakurai is aware people want Ashley because she also had one. I have a feeling time constraints might have been a factor in why she is not on the main roster. if she were to be DLC, do you think they'll replace her trophy with someone else?

Wigglytuff would make for a good surprise character (since Pichu is in the game) Igglybuff seems like it'd be lighter than Jigglypuff (who is already the lightest character in the game) and die earlier because of it. Not a good choice for a clone.
